The A65 between Skipton and Gargrave was closed in both directions on Saturday evening after a two-car crash near Thorlby.
Firefighters from Skipton freed a woman driver, in her thirties, who was trapped in her car. She was taken to hospital on a spinal board with suspected pelvic injuries.
Her female passenger, in her forties, suffered head injuries and was also taken to hospital.
A second car had overturned but the male driver was reported to be uninjured.
The road was closed from about 5.30pm until well into the evening.
Two fire appliances attended the crash and used hydraulic cutting equipment to free the trapped driver.
The road has now re-opened.
